ABSTRACT

Objective::To study the effect of total alkaloids of Nandina domestica in attenuating arsenic trioxide, and analyze chemical constituents of alkaloids extract of Nandina, in order to provide the theoretical basis for studying the effect of N. domestica in attenuating arsenic trioxide with alkaloids extract of N. domestica as effective fraction. Method::The model of acute liver injury induced by arsenic trioxide was used to compare the effects on heart and liver functions of mice between arsenic trioxide alone and total alkaloids of N. domestica combined with arsenic trioxide. The detoxification of total alkaloids on arsenic trioxide was evaluated based on biochemical parameters and pathological report. Peakview (Version1.2, AB SCIEX) software was used to process the mass spectrometry data of total alkaloids of N. domestica. The structures of determined compounds were identified by molecular weight of compound (molecular formula), secondary fragments, chromatographic peak retention and literature information. Result::Among biochemical indicators, creatine kinase(CK), lactate dehydrogenase(LDH), blood urea nitrogen(BUN), malondialdehyde(MDA), alanine aminotransferase(ALT) and amino transferase of aspartate(AST) of the arsenic trioxide group were increased, while elimination rates of Na+ -K+ -adenosine triphosphate(ATP), superoxide dismutase(SOD), catalase(CAT) and serum creatinine(SCr) were decreased, compared with those of the combination group. CK and LDH of the alkaloids extract group were more obviously increased than those of the N. domestica extract group, but with no remarkable difference. In histomorphometric examination, edema of mouse heart cells was improved, and some kidney and liver damages in rats were alleviated. Totally 25 alkaloids of alkaloid extract were identified. Among them, 18 were known, and 7 were unknown, including 3 structural types, in which apomorphine alkaloids were mostly. Conclusion::Heart, kidney and liver damage degrees of the combination group were significantly alleviated compared with the arsenic trioxide group. The total alkaloids fraction extracted and purified have a significant attenuation in arsenic trioxide toxicity. The detoxification of total alkaloids extract was equal to that of N. domestica extract. Furthermore, apomorphine alkaloids, such as nantenine and domesticine, can be used as index components to establish a quality control method for total alkaloids.

ABSTRACT

Objective To analyze the general characteristics of voluntary blood donors, and to provide the basis for establishing effective recruitment mechanism. Methods The information of all whole blood donors (including mobile blood collection cars on the streets , blood donation houses and team blood donors) from 2006 to 2015 was obtained from the Zhejiang blood management information system. The sex, age, occupation, weight, and the amount of blood donation at a time of blood donors were descriptively analyzed. Results There were 5299700 frequencies of donating blood in Zhejiang Province from 2006 to 2015. The annual frequency was on the rise, with an average annual growth rate of 2.99%. There were 3098500 frequencies of male and 2201300 of female with the ratio of 1.4 :1. The frequency of female has declined since 2010. The majority of voluntary blood donors ranged from 18 to 45 years old, and the percentage was 91.44% . The ratios of staff, students and workers were 14.61%, 12.27%, 10.09%, respectively. The annual frequency of staff was on the rise, with an average annual growth rate of 7.71%. The frequency of workers was on the decline, and the frequency of students fluctuated over the years. The ratio of blood donation 400 mL at a time was 41.23%. For 400 mL blood donation at a time, the ratio of male donors (48.86%) was significantly higher than the female donors (30.50%, P<0.001) . In addition, the ratio of blood donation 400 mL at a time was proportional to age and body weight (P<0.001) . Conclusion In Zhejiang Province, the frequency of blood donation has increased year by year. Staff, students and workers with 18 to 45 years old are the main force of blood donation. The frequency of female blood donation has a tendency of dropping, and the ratio of blood donation 400 mL at a time is relatively low. It's necessary to strengthen the propaganda of blood donation.

ABSTRACT

Molecular imprinting technology is widely used in the separation and analysis of compounds such as flavonoids, alkaloids and polyphenols, due to its high selectivity and specific recognition and so on. However, no much of attention has been paid to the terpenoids. This paper is aimed to not only review the effects of common synthetic elements such as functional monomers, cross-linking agents and porogens on the polymer properties, but also highlight the application of terpene molecular imprinting in solid phase extraction, sensor, membrane separation and chromatographic separation by means of statistical analysis of literature. Furthermore, the shortcomings and improvement directions are discussed. We believed that this paper could provide references for better applications of molecular imprinting techniques to the analysis of terpenoid compounds.

ABSTRACT

AIM To study the dynamic changes of color and five constituents during processing of Gardeniae Fructus.METHODS Camera and Adobe Photoshop software were applied to collecting images and obtaining chromatic values (L,a,b) of processed products, respectively.HPLC was adopted in the content determination of genipin-1-β-D-gentiobioside,geniposide,crocin-Ⅰ,crocin-Ⅱ,crocin-Ⅲ.Then the correlation between color and constituent contents was investigated by linear regression and partial least squares method.RESULTS With the process of processing, the chromatic values of epicarp and seed pellets expelled showed decreasing trends, especially for that of the former. On the whole, the contents of geniposide,crocin-Ⅰ and crocin-Ⅱ were decreased, while those of genipin-1-β-D-gentiobioside and crocin-Ⅲ were first increased and then decreased. The chromatic values of epicarp and seed pellets expelled showed significant correlations with crocin-Ⅰ content.CONCLUSION Color and crocin-Ⅰ can be considered as control index and monitoring index during processing of Gardeniae Fructus, respectively.

ABSTRACT

<p><b>BACKGROUND</b>Glutamic acid decarboxylase antibody (GADA) and protein tyrosine phosphatase antibody (IA-2A) are two major autoantibodies, which exert important roles in the process of type 1 diabetes mellitus (T1D). Our study aimed to investigate the changes in positivity and titers of GADA and IA-2A during the course of Chinese acute-onset T1D patients and their relationships with clinical features.</p><p><b>METHODS</b>Two hundreds and forty-seven Chinese newly diagnosed acute-onset T1D patients were consecutively recruited. GADA and IA-2A were detected at the time of diagnosis, one year later, 3-5 years later after diagnosis during the follow-up; all the clinical data were recorded and analyzed as well.</p><p><b>RESULTS</b>During the course of acute-onset T1D, the majority of patients remained stable for GADA or IA-2A, however, a few patients changed from positivity to negativity and fewer patients converted from negativity to positivity. The prevalence of GADA was 56.3% at diagnosis, decreasing to 50.5% one year later, and 43.3% 3-5 years later while the corresponding prevalence of IA-2A were 32.8%, 31.0% and 23.3%, respectively. The median GADA titers were 0.0825 at diagnosis, declining to 0.0585 one year later and 0.0383 3-5 years later (P < 0.001), while the corresponding median titers were 0.0016, 0.0010, 0.0014 for IA-2A, respectively. Fasting C-peptide (FCP) and postprandial C-peptide 2 hours (PCP2h) levels of GADA or IA-2A negativity persistence patients were higher than those of positivity persistence and negativity conversion patients (P < 0.05) which indicated GADA or IA-2A negativity persistence T1D patients had a less loss of β cell function.</p><p><b>CONCLUSION</b>Our data suggest that repeated detection of GADA and IA-2A are necessary for differential diagnosis of autoimmune diabetes and the indirect prediction of the β cell function in Chinese patients.</p>
